Test Drive: ELISA Workflow

1) Create ELISA Experiment
  • Go to Projects App 
  • Click the ELISA project
  • Click + Experiment
  • Select the ELISA Binding Condensed Workflow 
  • Enter a unique Name for the Experiment, i.e., your initials - ELISA Binding - 1
  •  Note: You may enter a description, apply tags, or schedule a task (doing so will add it to the calendar in the Scheduling app), but it is not required 
  • Click Next
  • Click Add New Entities 
  • Enter 3 for the Number of Entities 
  •  Note: You may enter values in each field, but it is not required 
  • Click Next
  • Click Save & Submit 
  • Click Submit Entities to send the Entities/samples to LIMS for processing
2) Create LIMS Worksheet
  • Go to LIMS App 
  •  Note: On the left you will see the LIMS backlog of queued Entities. By default the LIMS backlog will group Entities by Experiment. 
  •  Note: Click the arrow next to your queued Experiment to expand and view the associated Entities.
  • Click the blue create worksheet icon to the right of the Experiment name in the LIMS backlog
  • Click Add Worksheet
3) Perform ELISA Experiment
  • Click the name of the Worksheet you just created
  • ----- Protocol 1: Plate Coating and Antibody Selection -----
  • Click the Flexview icon (located to the left of the "1 of 4 Protocols" indicator) to ensure you're viewing the Protocol is a document view
  •   Note: Some information is already populated (i.e. Instrument info)
  • Select the Consumable reagents for 1.2: Plate Coating
  •  Note: After selecting a consumable reagent, information about that item is pulled into read-only fields 
  •  Note: Barcode scanning can be setup instead of user selection
  • Enter a value of at least 1 for each Consume field
  • Click the Consume button to apply the value to Total Consumed 
  • Repeat for 1.3: Standards
  • Select a Reference Standard from the picklist
  • Repeat for 1.4: Controls
  • Select Plate Map A  
  • Click Save 
  • Click Plate Aliquots 
  •  Note: This creates a plate and replicate samples to populate the plate using the samples, standards, and controls selected in this Protocol.
  •  Note: Once created, the Plate's name and link will include the date/time of creation
  • Click Sample and Control Location Link to review the plate and sample placement
  •  Note: A new tab will open showing your 96-Well Plate details page (Assets app > Containers), displaying the layout of samples, standards, and controls
  • Return to your LIMS Worksheet tab
  • Click Save to continue
  • ----- Protocol 2: Softmax Settings -----
  •  Note: This Protocol is to confirm the instrument settings, no action is required
  • Click Save to continue
  • ----- Protocol 3: QC ELISA Quant Results - Binding -----
  • Click Parse Softmax to simulate an integration that will pull in the data files and generate a Plotly chart
  •  Note: The chart is interactive, allowing you to zoom in and inspect your data
  •  Optionally, click on any of the attached Instrument Output Files to view its contents
  •  Note: Clicking an attached file will redirect you to the Data app, where uploaded files are managed in L7|ESP
  • Click Save to continue
  • ----- Protocol 4: QC ELISA Concentration -----
  •  Note: This is a review of the final data
  • Sign off on the results, enter your Username and Password for the Approved By field
  • Click Sign to approval
  • Click Save to complete the Experiment
